In fluorescence analyses, it is essential that the light source be segregated from the emitted spectrum from the fluorophore so as not to interfere with the analyses. In a dichroic filter, the excitation wavelengths are highly reflected while the emission spectrum is transmitted. Both these rays are incident at 45o.
